ABSTRACT

5-aminovalanoic acid (5AVA) can be used as the precursor of new plastics nylon 5 and nylon 56, and is a promising platform compound for the synthesis of polyimides. At present, the biosynthesis of 5-aminovalanoic acid generally is of low yield, complex synthesis process and high cost, which hampers large-scale industrial production. In order to achieve efficient biosynthesis of 5AVA, we developed a new pathway mediated by 2-keto-6-aminohexanoate. By combinatory expression of L-lysine α-oxidase from Scomber japonicus, α-ketoacid decarcarboxylase from Lactococcus lactis and aldehyde dehydrogenase from Escherichia coli, the synthesis of 5AVA from L-lysine in Escherichia coli was achieved. Under the initial conditions of glucose concentration of 55 g/L and lysine hydrochloride of 40 g/L, the final consumption of 158 g/L glucose and 144 g/L lysine hydrochloride, feeding batch fermentation to produce 57.52 g/L of 5AVA, and the molar yield is 0.62 mol/mol. The new 5AVA biosynthetic pathway does not require ethanol and H2O2, and achieved a higher production efficiency as compared to the previously reported Bio-Chem hybrid pathway mediated by 2-keto-6-aminohexanoate.

ABSTRACT

A descorna cirúrgica a campo ainda e uma prática comum em animais de produção, apesar deste procedimento na maioria ainda se realizado por leigos, ou realizada em animais com menos de um ano de idade com ferro candente (avermelhado), esta conduta geralmente é efetuada na propriedade, sendo executada pelo próprio proprietário ou funcionário. O presente experimento usando anestesia geral e bloqueio local do nervo córneo e circularmente na base do corno com abraçadeira de naylon para sutura de pele, associada a ligadura da artéria e veia cornual mostrou ser eficiente reduzindo o tempo cirúrgico a campo e promovendo uma prevenção antecipada de hemorragia que é frequente para este procedimento.(AU)


The surgical dehorning the field and still a common practice in farm animals, although this procedure in most still held by lay people, or performed on animals less than one year old with red-hot iron (red), this conduct is usually done on the property, being executed by the owner himself or employee. This experiment using general anesthesia and local lock of corneal nerve and round the horn base with clamp naylon for skin suture, associated with ligature of the artery and vein cornual is efficient by reducing surgical time field and promoting an early prevention of bleeding is frequent for this procedure.(AU)


El quirúrgica descorne el campo quieto y una práctica común en los animales de granja, aunque este procedimiento en la mayoría todavía en manos de los laicos, o lleva a cabo en animales de menos de un año de edad con hierro al rojo vivo (rojo), este comportamiento se realiza generalmente en la propiedad, los trabajos realizados por el propietario o el propio empleado. Este experimento usando anestesia bloques general y local de los nervios de la córnea y alrededor de la base del cuerno con naylon pinza de sutura de la piel, asociados con la ligadura de la vena y la arteria cornual fue eficiente que reduce el tiempo quirúrgico el campo y la promoción de una prevención temprana sangrado que es común para este procedimiento.(AU)

ABSTRACT

O uso de novos materiais nanocompósitos para fabricação de dispositivos protéticos, matrizes para preenchimento de defeitos ósseos, proteção do tecido pulpar de dentes e túbulos dentinários expostos pode colaborar significativamente na qualidade de vida da população. Objetivo: Realizar o processamento da matriz polimérica de poliamida-6 (P6) impregnada com nanopartículas de trimetafosfato (TMP) e nanopartícula de prata (AgNP), avaliar a atividade antimicrobiana contra Streptococcus mutans e Candida albicans e a liberação de TMP e Ag+ . Métodos: Foi determinado a concentração inibitória mínima (CIM) da AgNP com ou sem a presença da NH3 para S. mutans e C. albicans. Em seguida, realizado a síntese e caracterização dos nanocompósios (P6, P6-2,5, 5 e 10%TMP associado ou não a AgNP), quantificação das unidades formadoras de colônia (UFC), determinação do halo de inibição e quantificação de TMP e Ag+ liberados durante 1, 2, 3, 4, 5, 10, 12, 14, 16, 18, 20 e 24 horas em água deionizada. Os dados foram submetidos à análise de variância bidirecional, seguida do teste de Fisher LSD (p< 0,05). Resultados: A CIM para C. albicans foi de 9,40 mg/mL com ou sem a presença da NH3 e para o S. mutans foi de 601,9 mg/mL com NH3 e 300,9 mg/mL sem NH3. Nos testes de caracterização foi possível incorporar a P6 com TMP sem alterar suas propriedades. A maior quantidade de Ag+ liberada ocorreu nas primeiras três hora para todos os grupos decorados com AgNP. Houve liberação de TMP nas primeira três horas para os grupos P6-5%TMP e P6-10%TMP e para as demais membranas não foram detectadas liberações. No ensaio de difusão em ágar os halos formados para C. albicans e S. mutans mostraram ação da AgNP para ambos os microrganismos. Os grupos P6-Ag2,5%TMP e P6-Ag-5%TMP com AgNP apresentam maior redução de UFC para S. mutans quando comparado aos demais grupos, com maior redução no tempo de 18 horas. Para C. albicans todos os grupos apresentaram redução na UFC quando comparado ao controle, sem diferença estatística entre os mesmo. Conclusão: Foi possível desenvolver um matriz polimérica de P6 impregnada com TMP e AgNP com ação antimicrobiana contra os microrganismos testados(AU)


The use of new nanocomposite materials for the manufacture of prosthetic devices, matrices for filling bone defects, protection of the pulp tissue of exposed teeth and dentinal tubules can significantly contribute to the quality of life of the population. Objective: To perform the processing of the polymeric matrix of polyamide. 6 (P6) impregnated with trimetaphosphate nanoparticles (TMP) and silver nanoparticles (AgNP), evaluate the antimicrobial activity against Streptococcus mutans and Candida albicans and the release of TMP and Ag +. Methods: The minimum inhibitory concentration (MIC) of AgNP was determined with or without the presence of NH3 for S. mutans and C. albicans. Then, the synthesis and characterization of the nanocomposites (P6, P6-2.5, 5 and 10% TMP associated or not with AgNP), quantification of colony forming units (CFU), determination of the inhibition halo and quantification of TMP and Ag + released during 1, 2, 3, 4, 5, 10, 12, 14, 16, 18, 20 and 24 hours in deionized water. The data were submitted to bidirectional analysis of variance, followed by the Fisher LSD test (p < 0.05). Results: The MIC for C. albicans was 9.40 mg / mL with or without the presence of NH3 and for S. mutans it was 601.9 mg / mL with NH3 and 300.9 mg / mL without NH3. In the characterization tests it was possible to incorporate the P6 with TMP without changing its properties. The highest amount of Ag + released occurred in the first three hours for all groups decorated with AgNP. There was TMP release in the first three hours for the P6-5% TMP and P6-10% TMP groups and for the other membranes, no releases were detected. In the agar diffusion assay, halos formed for C. albicans and S. mutans showed AgNP action for both microorganisms. The P6-Ag-2.5% TMP and P6-Ag-5% TMP groups with AgNP show a greater reduction in CFU for S. mutans when compared to the other groups, with a greater reduction in the time of 18 hours. For C. albicans, all groups showed a reduction in CFU when compared to the control, with no statistical difference between them. Conclusion: It was possible to develop a polymeric matrix of P6 impregnated with TMP and AgNP with antimicrobial action against the microorganisms tested(AU)

Abstract Denture adhesives need complete removal due to their frequent replacement. Objective Our study investigates the removal of denture adhesives from denture base materials, using different methods. Methodology PMMA and Polyamide denture base materials were used to fabricate 120 samples (15×15×1.5mm). One side of the samples was left as processed and the other polished with a usual procedure, hydrated for 24 h, dried, and weighted. They received 0.2 g of three adhesive creams on their unpolished surface (Corega, Olivafix, Fittydent), pressed on polysulfide material, stored under 37°C and 95% rel. humidity for 1 h and 60 of them, following their separation from polysulfide base, brushed under running water, whereas the rest inserted in a cleanser bath (Fittydent Super) for 5 min. The samples were dried and inserted in the oven (37°C) for additional 10 min and weighted again. Roughness tests of denture materials and light microscopy of adhesives creams were also used to evaluate the materials. Time lapse images of spayed with water adhesives on PMMA base were also taken to evaluate the volumetric changes of adhesives. Weight data before and after adhesive removal, indicating the amount of remaining adhesive, were statistically analyzed using Welch's ANOVA and Games-Howell multiple comparisons tests at α=0.05 level of significance. Results Roughness of Polyamide was higher than PMMA and Fittydent showed greater volumetric changes than the others. Significant differences (p<0.05), were found between PMMA and Polyamide bases, between Olivafix and Fittydent adhesives, and between brushing and cleansing methods but only for PMMA-Olivafix combination. Conclusions Adhesives showed a stronger adherence to PMMA surface, and Fittydent was the most difficult to be removed. Removal methods were not effective for all adhesives or denture base materials. These indicate that removal methods, adhesive type and denture base material are all playing a significant role in the removal of adhesives from denture surfaces.

RESUMEN La investigación se realizó a fin de evaluar el efecto del gas de formalina sobre el recuento de mesófilos en nailon comercial (poliamida) destinado a procedimientos quirúrgicos. En primer lugar, se evaluó la contaminación de una muestra del material comercial de 1 g expuesta al ambiente de quirófano por 72 horas, a través de la técnica de recuento en placa para mesófilos; se obtuvieron 850 UFC/g. Una vez comprobada la contaminación de la poliamida comercial, esta se sometió a gases de formalina en comprimidos. Se colocaron 5 muestras (n=5) de nailon de 1 g cada una en 5 recipientes herméticos de 1 litro con 1 gramo de formalina cada uno; estos recipientes se almacenaron en un mueble en sala de esterilización a un metro de altura del piso y posteriormente, fueron abiertos y cultivados a través de la técnica de recuento en placa para mesófilos, uno por día a lo largo de 5 días a intervalos de 24 horas. Los resultados obtenidos no registraron crecimientos de microorganismos a partir de las 24 horas y durante los 5 días posteriores y se encontraron diferencias estadísticamente significativas (p < 0,05). Se concluye que bajo las condiciones del presente estudio el tiempo de esterilización de la formalina sobre nailon comercial es de 24 horas.


ABSTRACT Hie present research work was carried out with the purpose of evaluating the effect of formalin gas on the count of mesophiles in commercial nylon (polyamide) destined to surgical procedures. Firstly, the contamination of a 1 g commercial nylon sample, exposed to the operating room environment for 72 hours, was evaluated through the plate counting technique for mesophiles; it yielded a result of 850 CFU / gram. Once the contamination of the commercial polyamide was checked, it was subjected to formalin gases in tablets. Five nylon samples (n=5) of 1 gram each were placed in 5 1 liter airtight containers containing 1 gram of formalin; Hese containers were stored in a cabinet in the sterilization room one meter above the floor, and later opened and cultivated through the plate counting technique for mesophiles, one per day for 5 days at 24 hour intervals. The results obtained after being exposed to formalin gases in tablets did not register growths of microorganisms after 24 hours and during the 5 days after the study, finding statistically significant differences (p < 0.05) in relation to the studied times concluding under the conditions of the present study that the sterilization time of the formalin on commercial nylon is equal to 24 hours.

ABSTRACT

The aim of this study was to develop an experimentally-induced canine model of left ventricular hypertrophy through banding of the ascending aorta using nylon ties. Seven clinically normal dogs free of cardiovascular disease were used. Nylon tie was used in banding the mid-ascending aorta. Clinical, radiographic and echocardiographic evaluations were done at 1.5, 3 and 6 months. Dogs were euthanized at 6 months for post mortem and histopathological evaluation. Clinically, dogs did not exhibit any signs of cardiovascular disease at 1.5 or 3 months, while at 6 months two dogs (28.6 %) exhibited mild weight loss, exercise intolerance and heart murmurs. Radiographic evaluation revealed significant increase in cardiac size only at 6 months based on measurement of the cardiothoracic area evaluation. Echocardiography revealed increased left ventricular wall thickness starting from 1.5 month, although this increase was statistically significant at 3 and 6 months (p > 0.05). Left ventricular hypertrophy was confirmed by post mortem examination. Histopathological sections of left ventricle in all dogs demonstrated myocyte hypertrophy and interstitial fibrosis. This model simulates the naturally occurring ventricular hypertrophy using a rapid and economic technique. Such models are required to understand pathogenesis of heart disease and to develop effective treatment strategy.

ABSTRACT

Polyamide-amine (PAMAM) dendrimer, a new hyperbranched macromolecular polymer, is considered an "artificial protein" by many scholars on account of its excellent chemical and biological characteristics. PAMAM has internal cavities and a large number of reactive terminal groups. These structures allow the polymer to be used as a bionic macromoleculethat could simulate the biomimetic mineralization of the natural organic matrix on the surface of tooth tissue. Specifically, PAMAM can beused as an organic template to regulate mineral nucleation and crystal growth; thus, the polymerisa more ideal dental restoration material than traditional allogenic materials. This article reviews research progress on thePAMAM-induced biomimetic mineralization of hard tooth tissues.

RESUMEN: El objetivo de este estudio fue comparar los hilos de sutura de seda negra y la sutura nylon con respecto a la adhesión bacteriana post extracción dental simple de piezas posteriores. El presente trabajo se desarrolló en la Clínica Odontológica y el Laboratorio de Microbiología de la Unidad de Ciencias Biomédicas y Biotecnología de la Universidad Privada Antenor Orrego (Trujillo, Perú), entre agosto y setiembre de 2014. Se trabajó con 19 unidades de muestreo para hilo de seda negra y 19 para nylon. El procedimiento de exodoncia para todos los pacientes fue estandarizado. Luego de la extracción, se procedió a suturar la herida quirúrgica, colocando dos puntos circunferenciales, uno con cada tipo de hilo. A los 7 días, los hilos fueron retirados y sometidos a análisis microbiológico. La adhesión bacteriana de los hilos de sutura seda negra y nylon fue comparada empleando el test de Wilcoxon. Además la adhesión bacteriana de cada hilo fue comparada con la adhesión en hilos estériles (control) empleando el test de Mann-Whitney para una p<0,005. Al comparar la adhesión bacteriana de ambos hilos, se encontró diferencia entre ellos (p=0,027), correspondiendo mayor adhesión al hilo de nylon. La adhesión bacteriana fue mayor en el hilo de sutura de nylon comparado con el hilo de sutura de seda negra.


ABSTRACT: The objective of the study was to compare black silk suture threads and nylon suture with respect to bacterial adhesion after simple dental extraction of posterior pieces. The present study was developed in the Clínica Estomatológica and the Laboratorio de Microbiología of the Unidad de Ciencias Biomédicas y Biotecnología de la Universidad Privada Antenor Orrego (Trujillo, Peru), between August and September 2014. We worked with 19 units sampling for black silk thread and 19 for nylon. The procedure of exodontia for all patients was standardized. After the extraction, the surgical wound was sutured, placing two circumferential points, one with each type of thread. After 7 days, the threads were removed and subjected to microbiological analysis. The bacterial adhesion of the black silk and nylon suture strands was compared using the Wilcoxon test. In addition, the bacterial adhesion of each strand was compared with the adhesion in sterile strands (control) using the Mann-Whitney test for p <0.005. When comparing the bacterial adhesion of both threads, a difference was found between them (p = 0.027), corresponding greater adhesion to the nylon thread. Bacterial adhesion was higher in nylon suture compared to black silk suture.

RESUMO Objetivo: avaliar o perfil de segurança e os resultados estéticos do 2-octilcianoacrilato versus sutura intradérmica com fio de nylon em cirurgias mamárias. Métodos: ensaio clínico randomizado, aberto, que avaliou a ocorrência de complicações, como deiscência, hematoma, infecção e reações alérgicas após o uso do 2-octilcianoacrilato ou do fio de nylon. Também foi analisado o tamanho das incisões, o tempo de fechamento da pele e o tempo cirúrgico total. O resultado estético foi avaliado após 40 e 180 dias da cirurgia, por meio da largura média da ferida operatória e por avaliação subjetiva conceitual (ótimo, bom, razoável ou ruim). Resultados: foram incluídas 79 pacientes, sendo 37 no grupo 2-octilcianoacrilato e 42 no grupo de sutura com fio de nylon. O estudo foi interrompido antes do término do recrutamento dos pacientes pela ocorrência de maior número de deiscências no grupo do adesivo (OR: 11,42; IC95%: 1,36-96,02; p=0,007). Em relação às demais complicações analisadas, ao tempo cirúrgico e ao resultado estético no pós-operatório, não se observaram diferenças significativas entre os grupos. A média do tamanho da ferida operatória foi maior no grupo do adesivo em relação ao grupo da sutura, mas não houve correlação entre o tamanho da ferida e o maior número de deiscências. Conclusão: o 2-octilcianoacrilato apresentou maior risco de deiscência em relação à sutura intradérmica, com resultados estéticos equivalentes.


ABSTRACT Objective: to evaluate the safety profile and aesthetic results of 2-octyl-cyanoacrylate versus intradermal nylon suture in breast surgeries. Methods: an open-label, randomized, clinical trial evaluating the occurrence of complications, such as dehiscence, hematoma, infection, and allergic reactions after the use of 2-octyl-cyanoacrylate or nylon thread. The size of the incisions, skin closure time, and total surgical time were also analyzed. The aesthetic outcome was evaluated at 40 and 180 days after surgery, by means of the average width of the surgical wound and by subjective conceptual assessment (optimal, good, reasonable, or poor). Results: 79 patients were included: 37 in the 2-octyl-cyanoacrylate group and 42 in the nylon suture group. The study was stopped before the end of patient recruitment due to the occurrence of a greater number of dehiscences in the adhesive group (OR: 11.42; 95%CI: 1.36-96.02; p=0.007). Regarding the other analyzed complications, the surgical duration and postoperative aesthetic result, no significant differences were observed between the groups. The mean operative wound size was greater in the adhesive group than in the suture group, but there was no correlation between wound size and the largest number of dehiscences. Conclusion: while the cosmetic outcomes with the two techniques were similar, there was a greater risk of dehiscence with the use of 2-octyl-cyanoacrylate compared to intradermal suturing.

ABSTRACT

PURPOSE: We report a case of utilizing a previous silicone band track in the reoperation of scleral encircling. CASE SUMMARY: An 8-year-old male presented with rhegmatogenous retinal detachment in the right eye. Five days after this diagnosis, he received scleral buckling surgery and cryopexy to seal the retinal tear. One month after surgery, a fundus examination showed subretinal fluid at the inferior site of the scleral buckle. He underwent scleral encircling surgery and a cryopexy procedure. The patient has had an uneventful postoperative course, and the retina has remained attached over a follow-up period of 9 months. However, exotropia and hypotropia developed in the right eye. Diagnosis of restrictive strabismus due to tissue adhesion around the silicone band was made. The encircling band was therefore removed and laser photocoagulation was performed 360° around the retina. Twenty-four hours after surgery, a fundus examination showed subretinal fluid. He received 360° scleral encircling surgery not using the 360° conjunctival peritomy. After confirming a previous encircling tract using #0-0 polydioxanone as a guide, #5-0 Nylon was tied to the end of the guide and inserted through the encircling tract with the end sutured with the silicone band. The silicone band was inserted into the encircling tract by pulling the #5-0 Nylon as a guide. Ophthalmoscopy revealed an attached retina with indentation of the scleral buckle at 360°. CONCLUSIONS: For reoperation in patients who previously underwent scleral encircling surgery, using the previous scleral encircling tract may be effective in cases with conjunctival and tissue adhesion.

ABSTRACT

PURPOSE: Surface alterations of titanium discs following instrumentation with either a nylon brush or a metal brush were evaluated. METHODS: A total of 27 titanium discs with 3 surface types (9 discs for each type), including machined (M) surfaces, sandblasted and acid-etched (SA) surfaces, and surfaces treated by resorbable blast media (RBM), were used. Three discs were instrumented with a nylon brush, another 3 discs were instrumented with a metal brush, and the remaining 3 discs were used as controls for each surface type. Surface properties including the arithmetic mean value of a linear profile (Ra), maximum height of a linear profile (Rz), skewness of the assessed linear profile (Rsk), arithmetic mean height of a surface (Sa), maximum height of a surface (Sz), developed interfacial area ratio (Sdr), skewness of a surface profile (Ssk), and kurtosis of a surface profile (Sku) were measured using confocal microscopy. RESULTS: Instrumentation with the nylon brush increased the Ra, Sa, and Sdr of the M surfaces. On the SA surfaces, Ra, Sa and Sdr decreased after nylon brush use. Meanwhile, the roughness of the RBM surface was not affected by the nylon brush. The use of the metal brush also increased the Ra, Sa, and Sdr of the M surface; however, the increase in Sdr was not statistically significant (P=0.119). The decreases in the Rz, Sz, Ra, Sa, and Sdr of the SA surfaces were remarkable. On the RBM surfaces, the use of the metal brush did not cause changes in Ra and Sa, whereas Rz, Sz, and Sdr were reduced. CONCLUSIONS: Titanium surfaces were altered when instrumented either with a nylon brush or a metal brush. Hence, it is recommended that nylon or metal brushes be used with caution in order to avoid damaging the implant fixture/abutment surface.

ABSTRACT

PURPOSE: The purpose of this study was to compare mechanical and physical properties of injection-molded thermoplastic denture base resins. MATERIALS AND METHODS: In this study, six commercially available products (VA; Valplast, LC; Lucitone, ST; Smiltone, ES; Estheshot-Bright, AC; Acrytone, WE; Weldenz) were selected from four types of thermoplastic denture base materials (Polyamide, Polyester, Acrylic resin and Polypropylene). The flexural properties and shore D hardness have been investigated and water sorption and solubility, and color stability have evaluated. RESULTS: For the flexural modulus value, ES showed the highest value and WE showed significantly lower value than all other groups (P < .05). Most of experimental groups showed weak color stability beyond the clinically acceptable range. CONCLUSION: Within the limits of this study, thermoplastic denture base resin did not show sufficient modulus to function as a denture base. In addition, all resins showed discoloration with clinical significance, and especially polyamides showed the lowest color stability.

ABSTRACT

PURPOSE: To investigate the outflow characteristics of silicone tubes with intraluminal stents used in membrane-tube (MT) type glaucoma shunt devices. METHODS: The silicone tubes used in MicroMT (internal diameter of 100 µm with a 7-0 nylon intraluminal stent) and Finetube MT (internal diameter of 200 µm with a 5-0 nylon intraluminal stent) were connected to a syringe-pump that delivered a continuous flow of distilled water at flow rates of 2, 5, 10, and 25 µL/min. The pressures and resistances of tubes were measured at a steady flow rate with full-length, half-length, and absence of intraluminal stents. RESULTS: The mean outflow resistance of the two types of tubes ranged from 3.0 ± 1.9 to 3.8 ± 1.7 mmHg/µL/min with a full-length intraluminal stent, 1.8 ± 1.1 to 2.2 ± 1.1 mmHg/µL/min with a half-length intraluminal stent, and 0.1 ± 0.0 to 0.2 ± 0.0 mmHg/µL/min without an intraluminal stent. Theoretically, for a physiologic state with a flow rate of 2 µL/min and episcleral venous pressure of 6 mmHg, the mean pressures of tubes were expected to be 13.2 ± 3.0, 10.5 ± 2.4, and 6.4 ± 0.2 mmHg in MicroMT with full-length, half-length, and absence of intraluminal stents, respectively, and 12.5 ± 3.9, 9.6 ± 2.4, and 6.2 ± 0.2 mmHg in Finetube MT with full-length, half-length, and absence of intraluminal stents, respectively. The pressure variance also decreased with intraluminal stent retraction (p < 0.01). CONCLUSIONS: The small diameter tubes of 100 and 200 µm internal diameters, with 7-0 and 5-0 nylon intraluminal stents, respectively, used in the MT-type glaucoma shunt device showed safe and effective outflow characteristics.

ABSTRACT

Abstract Purpose: To compare two suture threads, poliglecaprone 25 and nylon, used as intradermal suture for skin closure in women undergoing their first cesarean section. Methods: This is a randomized clinical trial. A total of 60 women undergoing their first cesarean section were enrolled and prospectively assessed. They were randomly allocated to group I (n=30), which received an intradermal suture with nylon 4.0 or to group II (n=30), which had an intradermal suture with poliglecaprone 25, 4.0. The main author took standardized photographs of the scar 6 months after the operation. Four independent raters, two senior obstetricians and two senior plastic surgeons (a male and a female physician from each specialty) assessed the photographs.The panelists rated the scar according to Trimbos scale, composed by the subscales hypertrophy, color and width of the scar. Results: At baseline, patients in both groups were similar regarding age and body mass index. Five patients withdraw the study, four from group and one from group II. Scars of patients from group II were significantly less hypertrophic (p=0.001), thinner (p=0.019) and had more acceptable color (p=0.019). Conclusion: The intradermal suture with poliglecaprone 25 for skin closure after cesarean incision provides better aesthetic result.

ABSTRACT

Technological and tissue engineering have enabled available, biologically inert, and low cost materials to be considered as viable alternatives in the surgical treatment of long bone fractures in birds. The aim of this study was to microscopically analyse osteotomized humerus of birds following the insertion of solid laser-sintered polyamide 12 rods in order to detect foreign body reaction and, thus, verify the bioinert property of the material in the bone fracture environment. Polyamide 12 intramedullary rods were inserted into the osteotomized humerus of 10 birds (white Plymouth Rock) and blocked using 2mm diameter cortical screws of varying lengths. The birds were operated at 60 days of age and monitored post-operatively for three months. Animals were euthanized at 150 days old and samples of the operated humerus collected for immunohistochemistry, light and scanning electron microscopy analysis. Results show bone consolidation without rejection of the implant and absence of inflammatory cells. Vascular Endothelial Growth Factor (VEGF) was expressed in the endothelial cells of the blood vessels at the site of the newly formed bone surrounding the implant, indicative of local angiogenesis. There was no bone growth on the surface of the rod; however, the implant did not interfere with the circumjacent bone repair. Thus, the findings of this study corroborate with the literature in characterizing polyamide as a bioinert material and, under the studied conditions, it can be concluded that polyamide 12 intramedullary rod is biocompatible and provides adequate bone consolidation in humeral fractures with no signs of rejection.(AU)


Com o desenvolvimento tecnológico e crescimento da engenharia de tecidos, o uso de materiais disponíveis, bioinertes e debaixo custo pode ser alternativa viável para o tratamento cirúrgico de fraturas em ossos longos nas aves. O objetivo do estudo foi realizar avaliação microscópica óssea após a implantação de haste maciça de poliamida, implantada em úmeros osteotomizados de galinhas para detectar reação do tipo corpo estranho, verificando a propriedade "bioinerte" do material no ambiente de fratura óssea. Foram utilizados 10 galos (Plymouth rock branca) e implantou-se a haste perfazendo o bloqueio das mesmas com parafusos corticais de 2 mm de diâmetro com comprimentos de acordo com a necessidade. As aves foram operadas aos 60 dias de vida e o acompanhamento pós-operatório ocorreu por três meses. Após a eutanásia, foram realizadas coletas do úmero operado (local de consolidação óssea) paraexame histopatológico, imuno-histoquímico e de microscopia eletrônica de varredura. Os resultados demonstraram consolidação óssea, sem presença de rejeição do material, com ausência de células inflamatórias. A neoformação óssea ao redor do implante expressou VEGF (fator de crescimento endotelial vascular) nas células endoteliais dos vasos sanguíneos caracterizando angiogênese no local. Na região de interface de tecido ósseo com a poliamida não foram observadas micro fraturas. Não houve crescimento ósseo na superfície da haste, porém a mesma não atrapalhou o reparo ósseo circunjacente. Assim, esse estudo corrobora com a literatura caracterizando a poliamida como um material bioinerte, e nas condições estudadas pode-se concluir que o uso da haste intramedular de poliamida 12 proporcionou consolidação óssea nesse modelo biológico nesse tipo de fratura, não havendo indícios de induzir rejeição.(AU)

OBJECTIVES@#To establish a novel method for the separation of sperm cells in mixed stain, and to evaluate its application value.@*METHODS@#Totally 40 mixed stain samples were collected from sexual assault cases. Sperm cells were separated by the conventional differential lysis method and the nylon membrane bushing separation technique, respectively. The DNA of sperm cells was extracted with the silicon membrane kit (Forensic DNA Extraction Kit for Soft Tissues). The PCR amplification was performed using AmpFℓSTR® Identifiler® Plus kit, and the products were electrophoresed by 3500xL genetic analyser. The results of two separation methods were then compared.@*RESULTS@#Complete and single-source male STR genotypes could be obtained from all the 40 mixed stain samples except three samples with minimal residual of female DNA by the nylon membrane bushing separation technique. The STR genotypes of sperm cells could not be detected in 25 samples, which were obtained in 15 samples (seven were of incomplete male STR genotypes, six with residual of female DNA, two were complete and single-source STR genotypes of sperm cells).@*CONCLUSIONS@#The nylon membrane bushing separation technique developed in present study can be used in the separation of sperm cells in mixed stain, especially for the extraction of a small amount of sperm from a large quantity of female cells, which is inexpensive, rapid and simple.
